additional information Yarrowia lipolytica is engineered for enhanced production of arbutin. To construct the arbutin synthetic pathway in Yarrowia lipolytica, three genes, chorismate pyruvate-lyase (UbiC), 4-hydroxybenzoate 1-hydroxylase (MNX1), and hydroquinone glucosyltransferase (AS), are codon-optimized and heterologously expressed in Yarrowia lipolytica strain polf (leucine and uracil auxotroph). To maximize arbutin production, seven arbutin-biosynthesis molecular targets are overexpressed, and individual strengthening of DHS1 and DHS2 leads to an 8.9 and 7.8fold improvement in arbutin yield, respectively. Through optimization, a maximum arbutin titer of 8.6 g/l is achieved using the finally engineered Yarrowia lipolytica strain po1f-At0 Improvement in arbutin yields by glucose feeding Yarrowia lipolytica
